Measurement of chlorophyll meter under different conditions of temperature and light

The SPAD-502 Chlorophyll Meter is used to measure the color of the leaves, and guides crop fertilization. The pad value is a relative chlorophyll reading, also known as the green degree. Using pad values ​​to represent crop chlorophyll content has been applied to a variety of crops. Chlorophyll meter measured summer crops such as rice, cotton, soybean chlorophyll content and padd value to determine the correlation function of the padding value and chlorophyll content and found that two reached a very significant level, the correlation shows that the padding value can be used to represent the chlorophyll content Feasibility.

There was a very significant positive correlation between the paddle leaf values ​​of different wheat varieties and the chlorophyll content of the same variety, and other studies on the padding values ​​of rice, corn, sorghum and other crops also reached similar conclusions. At present, there is no report on the application of pad value in potato on chlorophyll meter. The warm light condition has a great influence on the padding value of the same leaf position. Lower temperature and shorter light hours are not conducive to the formation of potato chlorophyll. Longer light hours favor the formation of chlorophyll, but higher temperatures accelerate the decomposition of chlorophyll.

During the budding period of potato, healthy and non-invasive plants were selected in each treatment. The sap value of different leaf positions on the main stem was measured with a chlorophyll meter. Pour 2 leaves, pour 3 leaves until the bottom of a compound leaf. When the chlorophyll meter was used to measure the leaves of each leaf position, three different leaf parts were selected for measurement, and the average value was used as the spad value of the leaf. For each treatment, three plants were measured and the instrument measurement results were analyzed using SPSS software. Analysis of variance and multiple comparisons (SSR method).

Under different conditions of temperature and light, the variation of the padding value of different leaf positions of potato showed different rules. In the chlorophyll meter test set temperature and light treatment, the paddle value of the potato leaf was generally higher in the upper 4 leaves. The SPAD value decreased with the fall of leaf position. Under medium temperature conditions (light 12h and temperature 20°C), the change of the pad value between different leaf positions was small.
